Reynolds portrayed the character, a smart-mouthed mercenary, in X-Men Origins: Wolverine earlier in the year, but there had been rumours that he would not reprise the role after landing the lead in competing Warner Bros/DC Comics film Green Lantern.
Writing on his Twitter page, Liefeld relayed details from a meeting with X-Men and Deadpool producer Lauren Shuler Donner, saying: "Deadpool movie checklist - DP in costume-check! Breaking 4th wall-check! Loads of killing-double check! YES! Ryan Reynolds is on board!!!"
Reynolds earlier insisted that he can still play both roles, despite a potential conflct of interest.
