The Notebook actress had been rumoured for the role of Felicia Hardy, an ex-girlfriend of Spider-Man-turned-cat burglar, along with Glorious 39 star Romola Garai.
However, McAdams told Entertainment Weekly that the web speculation about her involvement in the movie was false.
"That's a total rumour, I have to say," McAdams commented. "I was hanging out in Toronto the other day and someone came up to me and said, 'I just heard you're doing Spider-Man 4'. And I said, 'Really? No-one told me!' It's not true."
Garai also ruled out her chances of appearing in the film, telling DS last week: "I had no idea that with Spider-Man 4 being such a big film... that people would pick up on it. I was a bit taken aback... I'm not going to get this [role]!"
