Speaking to Forbidden Planet, director Matthew Gratzner revealed that Larter is the frontrunner for the role of Virginia Lake.
"We've talked to Ali Larter, who's very interested," he said. "I've actually met with her, and I think she would be a great addition to the picture. Lake is very strong and obviously also very feminine, and Ali Larter definitely encompasses that."
Fringe star Joshua Jackson was recently cast in the lead role of Paul Foster.
UFO is based on a 1970s TV series of the same name.
Shooting is scheduled to start in the UK next spring.
