The fourth instalment in DreamWorks's animated franchise will be the last in the series and the first to be screened in 3D.
"We have always sought to open our Festival with films that are captivating and strike an emotional chord with movie-goers. Shrek Forever After combines the very best in storytelling and artistry while showcasing the wonders of innovative 3D filmmaking, [making it] one of the most anticipated new movies of 2010," said festival co-founder Jane Rosenthal.
DreamWorks Animation CEO Jeffrey Katzenberg said that it will be an "honour" to hold the movie's premiere at Tribeca, which runs from April 21 to May 2.
"It is a great privilege and tribute to our entire cast and crew to have been chosen as this year's opening night selection," he commented.
Mike Myers, Cameron Diaz and Eddie Murphy return to the voice cast for the new Shrek, which opens in cinemas this summer.
