According to Entertainment Weekly, the singer will join Cameron Diaz and Jason Segal as a substitute teacher who is the scion of a watch company fortune.
The film was written by the Office team of Lee Eisenberg and Gene Stupnitsky, and centres around an unorthodox middle school teacher.
Also joining the cast are Molly Shannon and Eric Stonestreet, who portrays Cameron on Modern Family.
Timberlake will next be seen in David Fincher's Facebook thriller The Social Network.
