According to The Hollywood Reporter, the studio has hired writer Chad St. John to pen the script, based on the 1980s adventure game from Midway.
Spy Hunter centres on a spy/assassin trained to eliminate rogue government agents.
Directors John Woo and Dwayne Johnson both tried to launch a Spy Hunter project at Universal, but those versions never got off the ground.
Warner apparently won the rights to the title when it bought Midway for $33m (£21m) last summer.
The studio has also tapped St. John to write the upcoming Sgt. Rock, based on the DC Comics character.
