According to Variety, Farmiga has landed a lead role in the independent production from Prominent Pictures.
The 36-year-old will play a preacher's wife looking for revenge.
Spanish director Daniel Calparsoro will helm the movie from a script by Russell Friedenberg.
A Thousand Guns is scheduled for a 2012 release.
Farmiga was recently seen in the Oscar-nominated Up in the Air.
