Spurlock will collaborate with Joss Whedon, Marvel Comics legend Stan Lee and Ain't It Cool News owner Harry Knowles for the project along with Legendary Pictures' Thomas Tull.
The film will follow seven fans who trek to the annual comic book and pop culture convention in San Diego. Producers are aiming for a theatrical release.
"When I was a kid, comics changed my life and my world. So when I got to San Diego, I felt like I was home," Spurlock said.
He added: "It was such an overwhelming experience for me, and I soon found out that I was not alone. So for me to get to share the experience of Comic-Con with fans all over the world will be an incredible opportunity."
Shooting is scheduled to wrap in July. Comic-Con takes place from July 22 to July 25.
