According to Total Film, the Oscar-winning actress could join Philip Seymour Hoffman in the project.
The story centres on a man called The Master (Hoffman) who leads a faith-based organisation similar to Scientology in the 1950s.
Jeremy Renner is also attached to co-star as someone who begins to question The Master's true intentions.
Witherspoon is said to be eyeing the role of The Master's pregnant wife Mary Sue.
Production is scheduled to begin in June.
