The actor will portray Dr. Abraham Erskine in the film, which will be released in July 2011, reveals The Hollywood Reporter.
The comics present Erskine as the scientist behind Project Rebirth, the clandestine experiment which inadvertantly creates Captain America.
Tucci joins Chris Evans, Hugo Weaving and Sebastian Stan, who have previously been confirmed.
Christopher Markus & Stephen McFeely wrote the script, which will be directed by Joe Johnston and released by Paramount Pictures.
