The Oscar-winning British director's survival drama will have its European premiere at the 54th annual festival on October 28. Boyle's Slumdog Millionaire closed the LFF in 2008.
127 Hours stars James Franco as Aron Ralston, the American mountain climber who was forced to amputate his own arm after getting trapped under a boulder in the Utah mountains in 2003.
"I am honoured and delighted that 127 Hours has been selected to close the BFI London Film Festival," Boyle said. "LFF played a vital role in the journey of Slumdog Millionaire in 2008 and it's great to be bringing new work here and renewing a happy partnership.
"I can't wait to unveil the new film and I hope it provides a worthy climax to what will hopefully be two weeks of great movies for our city."
The festival's artistic director Sandra Hebron called Boyle's latest project "bold and adventurous" and "a very fitting close to the festival".
127 Hours is due for a UK release in January 2011.
Carey Mulligan/Keira Knightley drama Never Let Me Go was earlier announced as the festival's opening night film.
The full LFF lineup will be unveiled on Wednesday, September 8.
