Bleeding Cool has reported that the Hellboy director was offered the chance to take on the relaunch, but has turned it down.
The news follows May's announcement that Del Toro would no longer direct the upcoming Hobbit films due to delays caused by MGM's financial problems.
The proposed reboot of the Superman series was announced following the lukewarm commercial reaction to Bryan Singer's Superman Returns in 2006.
Despite no official announcement from Warner Bros concerning the film's potential director, The Dark Knight's Christopher Nolan is attached to both produce and creatively guide the new film alongside David Goyer.
The Superman reboot is currently aimed to be released in late 2012.
