The period drama, starring Colin Firth, Geoffrey Rush and Helena Bonham Carter, revolves around King George VI hiring a speech therapist to help overcome a stutter. Tom Hooper's film held off competition from The First Grader to take home the 'People's Choice' award.
Audience prizes also went out to vampire film Stake Land and Sturla Gunnarsson's documentary Force Of Nature: The David Suzuki Movie.
Beautiful Boy, which sees Michael Sheen and Maria Bello play parents of a college student who goes on a killing spree, took home the 'International Critics' prize.
Previous winners of the Toronto audience award, which is often viewed as an early Oscars indicator, include Precious and Slumdog Millionaire.
