It was recently announced that Ifans has been cast as a villain in the Marc Webb-directed picture, but his character was not revealed.
Now The Wrap claims that the Welsh actor will play Dr Curt Connors, the scientist who turns into The Lizard after taking an experimental serum of reptilian DNA.
Connors was portrayed by Dylan Baker in the three Sam Raimi Spider-Man films, where the character was seen as Peter Parker's laboratory supervisor.
The report also states that Venom, Spider-Man's longtime nemesis, will not be featuring in the Sony picture.
Andrew Garfield and Emma Stone have been confirmed to star in the movie, which is due out in cinemas on July 3, 2012.
