The project, based on a web-published story by Max Barry, centres on an engineer at a technology firm who starts to replace his own body parts with robotic upgrades.
Aronofsky will reunite with his Black Swan writer Mark Heyman to adapt the "amped up pop-thriller" for the screen.
Barry has been publishing one page per day of the Machine Man manuscript on his website. The novel will be released by Vintage Books early next year.
Aronofsky will reportedly turn his attention to Machine Man after he completes comic book sequel Wolverine 2.
