The 70-year-old is in final negotiations to play the part of Uncle Ben in the Marc Webb-directed picture, according to The Hollywood Reporter.
The character of Uncle Ben famously inspires Peter Parker to become the web-slinging superhero following his death at the hands of a criminal. The role was played by Cliff Robertson in the Sam Raimi version of the comic book story.
It is also alleged that Forrest Gump actress Sally Field is in talks to play Aunt May, Parker's other close family member, although a deal is still thought to be some way off.
Columbia Pictures' Spider-Man, starring Andrew Garfield, Emma Stone and Rhys Ifans, is due out on July 3, 2012.
