The Oscar-winning actress, who previously worked with Pirates star Johnny Depp on Chocolat and director Rob Marshall on Nine, has a fleeting role as a noblewoman who gets robbed by Captain Jack Sparrow.
Dench will appear in a sequence at the start of the movie, which tracks Sparrow as he leaps across carriages on the streets of London to evade capture from King George II's guards.
The setpiece has been described as "Charlie Chaplin getting away from the Keystone Kops". Depp and Dench were said to be in "fits of giggles" during filming.
Penelope Cruz, Geoffrey Rush, Stephen Graham and Richard Griffiths co-star in On Stranger Tides, opening in 3D and conventional cinemas next year.
