According to The Playlist, Clooney is eyeing a reunion with director Steven Soderbergh on the spy remake. The pair previously worked together on Out Of Sight, the three Ocean's movies, Solaris and The Good German.
They also co-owned Section Eight, the production company behind Clooney's first two directorial efforts Confessions Of A Dangerous Mind and Good Night, And Good Luck.
Clooney's role has not been revealed, although he is likely to take on the part of lead spy Napoleon Solo. Robert Vaughn portrayed the character in NBC's original espionage show.
Warner Bros executives have allegedly approved Soderbergh's Man From U.N.C.L.E. storyline, which will shun a contemporary setting in favour of keeping the action in the '60s.
