According to Variety, the project centres on the relationship between a brain-damaged man and his sister.
Maya Forbes is penning the movie's script.
Brolin is also starring in the Coen Brothers' upcoming True Grit and the third Men in Black movie, which is currently in production.
The No Country for Old Men actor previously directed the short film X.
