Campbell Scott has already been signed to play Richard Parker, while Julianne Nicholson is currently in talks to play Mary Parker, according to The Hollywood Reporter.
Parker's parents have only made sporadic appearances in the comic series, following Peter's discovery that they were secret agents who were murdered shortly after his birth.
The report also suggests that Irrfan Khan and Annie Parisse are in negotiations to play a pair of super-villains.
The new Spider-Man film will be released in 3D on July 3, 2012.
