The project has been in development for over ten years, and was owned by troubled company MGM before Fox picked up the rights.
According to Empire, Fox will keep the Farrelly Brothers' original script with Mike Cerrone, but are to start again with many other elements of the proposed film.
During the early stages of the film's development, Sean Penn, Benicio Del Toro and Jim Carey were attached to star as the film's leads - however, it is currently unknown if this is still the case.
"We'll make it with the best possible people," Peter Farrelly said of the film. "It could be a major movie star or an unknown."
The Three Stooges is thought to begin shooting on March 14, 2011.
