The British actor screen-tested for the part earlier this month against Adrien Brody, Josh Lucas, James D'Arcy and Oliver Jackson-Cohen, and will now take on the eponymous role in Timur Bekmambetov and Tim Burton's 3D blockbuster.
The film is based on Seth Grahame-Smith's novel, which re-imagines the 16th President as a man on a quest to destroy all vampires during the Civil War.
Walker has appeared on screen in Kinsey and Flags Of Our Fathers. More recently he starred in the rock musical Bloody Bloody Andrew Jackson.
Abraham Lincoln: Vampire Hunter is due for release on June 22, 2012.
