According to a press release, Castle star Nathan Fillion will portray the title character alongside Mad Men's Elisabeth Moss.
Fillion said: "I was always reading the comic books, I was always a spectator. Now I feel like I own a little piece of Green Lantern."
Moss - who plays Peggy Olson in the AMC show - is to voice a new Lantern recruit named Arisia. Other actors cast in the film include Jason Isaacs as Sinestro, Henry Rollins as Kilowog and professional wrestler 'Rowdy' Roddy Piper as Bolphunga.
Released to promote this summer's live-action Green Lantern movie, the animated project is directed by Lauren Montgomery, Jay Oliva and Christopher Berkeley.
Green Lantern: Emerald Knights will be available on DVD, Blu-ray and download platforms from June 7.
