Leo, who won the corresponding prize at the Golden Globes for her role in The Fighter earlier this year, received the award from Kirk Douglas.
"I'm just shaking in my boots here," she said. "OK, alright, thanks David O. Russell, I want to thank the actors, Mark Wahlberg, Christian Bale, Amy 'my sweet sister' Adams."
Spotting the crowd in the gallery, she continued: "OK, yeah, I am kind of speechless. Golly, there's people up there too!"
She also mentioned previous winner Kate Winslet, saying that her acceptance speech two years ago for The Reader made it look "so f**king easy... oops!"
Fighting back tears, Leo also paid tribute to Alice Ward (whom her character is based on) and her "beautiful" family for "opening your hearts to all of us", becoming more emotional as she thanked her son.
She concluded by thanking the Academy of Motion Pictures and Sciences, saying: "It's about selling motion pictures and respecting all the work... thank you very much!"
