Warner Bros and Legendary Pictures have confirmed that the 46-year-old actress will play Martha Kent, Clark Kent's adoptive mother, in the forthcoming film.
In a press release, director Zack Snyder said: "This was a very important piece of casting for me, because Martha Kent is the woman whose values helped shape the man we know as Superman.
"We are thrilled to have Diane in the role because she can convey the wisdom and the wonder of a woman whose son has powers beyond her imagination."
Lane received an Oscar nomination for her role as Connie Summer in Unfaithful and recently starred in horse racing biopic Secretariat. She joins Henry Cavill, who has already been cast as Clark Kent.
Tentatively titled Superman: The Man of Steel, the Christopher Nolan-produced movie is due for release in December 2012.
