According to Black Film, Luke will play Gabe Jones in the film, known in the comic books for being one of Nick Fury's closest confidants.
The character is known for being the first African-American member of the group, and has remained a prominent character in the Marvel Universe since World War II by joining up with Fury's international peacekeeping force S.H.I.E.L.D.
It is not yet known whether Jones might also make an appearance in next year's team-up film The Avengers, although the character would be a lot older in the film's modern timeline and could conceivably be recast.
Luke is best known for his appearance in 2009 film Notorious, as well as in NBC's short-lived medical drama Trauma.
Captain America: The First Avenger is due for 3D release on July 22.
