The filmmaker and visual artist, whose work includes Blue Velvet, Mulholland Drive and Twin Peaks, is to oversee the Duran Duran webcast on March 23. The concert will be streamed live on YouTube at 10pm ET from Los Angeles' Mayan Theatre.
Fans will be invited to interact with the show via a 'Choose Your Cam' feature, 'Crowd Visualisers' and upload photos to be included in online collages.
"The idea is to try and create, on the fly, layers of images permeating Duran Duran on stage, a world of experimentation and hopefully some happy accidents," Lynch said.
Duran Duran keyboardist Nick Rhodes added to Billboard that he hopes the combination produces "mysterious, magical and surprising" results.
"The stranger it is, the more beautiful it will be," he commented. "What appeals to us very much about working with David is that we like the way he thinks about things and how he's always gone out on a limb to make things different than anybody else."
The Lynch/Duran Duran concert follows on from last year's Unstaged offerings from Arcade Fire and Terry Gilliam, John Legend and The Roots and the Kenny Ortega-directed Sugarland event.
