Speaking to Entertainment Weekly, the comedian admitted that he has not been approached to appear in the rumoured sequel.
"I read about that on the internet just like everybody else," said Hader, who stars in the new comedy Paul along with Simon Pegg and Nick Frost.
The Saturday Night Live cast member added: "I don't know where that [rumour] came from. I have not gotten a call. I don't know anything about the plot. I was very flattered to have people talking about me online in conjunction with that project.
"I love the Ghostbusters franchise and was so excited to hear there might be another one."
Hader said that, if asked, he would appear in Ghostbusters 3 "in a heartbeat".
