The Transformers actress is circling an unknown role in the untitled film, which revolves around Paul Rudd and Leslie Mann's characters from Apatow's 2007 box office hit Knocked Up.
Additionally, Deadline reports that Albert Brooks will board the cast as the father of Rudd's character Pete.
The comedy star has not appeared on the big screen since 2003's The In-Laws, but will soon be seen as the villain in Ryan Gosling's action thriller Drive.
Rudd and Mann's Knocked Up couple were loosely based on Apatow's own relationship with wife Mann. Apatow previously said that the comedy will not be a direct sequel to the earlier movie.
"It is just a story from Pete and Debbie's current life," he commented. "People really responded to their characters and problems."
