The 'Dances with Wolves' actor, who was first linked to the project last month, has agreed a deal to play Clark Kent's adoptive father Jonathan in the Warner Bros picture, according to Deadline.
Costner will be joined in the movie by confirmed cast members Henry Cavill, who is playing the titular superhero, and Diane Lane, who will portray Jonathan's wife Martha.
Costner was most recently seen starring alongside Ben Affleck and Tommy Lee Jones in John Wells's The Company Men.
Superman: Man of Steel is being produced by Batman trio Christopher Nolan, Emma Thomas and Charles Roven, as well as Zack Snyder's wife Deborah, from a script penned by David S. Goyer.
Other actors who have been rumoured for parts in the project include Rosamund Pike and Viggo Mortensen.
It is currently scheduled for release in December 2012.
