The project, based on a novel by Don Winslow, would see the actor take charge of a pair of drug-dealing hippies and ruthlessly force them into the employ of a tough drugs boss.
According to Deadline, Salma Hayek is also considering a part in the venture.
The script has been adapted by Shane Salerno, who has previously reworked hits such as Armageddon, Alien vs Predator, Shaft and Breakdown.
It is unknown who will take on the role of 'O', a girl who has a relationship with both of the leads.
Jennifer Lawrence, who has now committed to Hunger Games, was in the running for the part, the site claims.
Aaron Johnson and Taylor Kitsch are also signed up to the project, as the two main characters.
Del Toro is currently shooting his directorial debut, which constitutes a segment of the joint film 7 Days in Havana.
