The movie, to be directed by Morning Glory's Roger Michell, will be based on the BBC radio play by Richard Nelson.
It centres on the alleged affair between Roosevelt and his cousin Margaret Stuckley, which took place in June 1939 on the weekend that King George VI and the Queen visited the president at his New York cottage.
The meeting marked the first time the UK monarchy visited America.
Nelson will write the script for Hyde Park, while In The Loop's Kevin Loader is serving as a producer.
Murray is currently thought to be considering an offer to reprise his role as Peter Venkman in a third Ghostbusters movie.
