Gordon-Levitt will play John Blake, a Gotham City police officer assigned to special duty by Gary Oldman's Commissioner Gordon.
Oscar winner Cotillard has signed to portray Miranda Tate, a board member for Wayne Enterprises who encourages Bruce Wayne to continue his father's philanthropic ventures.
"When you collaborate with people as talented as Marion and Joe, it comes as no surprise that you would want to repeat the experience," said director Christopher Nolan, who worked with the actors in 2010 film Inception.
Nolan added: "I immediately thought of them for the roles of Miranda and Blake, and I am looking forward to working with both of them again."
Christian Bale, Anne Hathaway, Tom Hardy and Michael Caine will also star in the sequel to 2008's The Dark Knight.
Lost actor Nestor Carbonell has reportedly begun talks to reprise his role as the mayor of Gotham City in the project.
