The actor could be teaming up with Jake Gyllenhaal, who is in line for the second lead, Variety reports.
End of Watch will focus on the relationship between two long-time partners on the beat.
Harsh Times and Street Kings director David Ayer will be helming from his own script.
Pena will next be seen in the Universal comedy Tower Heist which co-stars Alan Alda and Ben Stiller.
He has previously taken on more serious roles in films including the domestic ensemble drama Crash, Battle: Los Angeles and The Shield.
