The Hollywood Reporter says that they will join the film's previously-confirmed leads Cameron Diaz, Colin Firth and Alan Rickman.
Joel and Ethan Coen have penned the script, which follows a British cat burglar's attempts to steal a priceless artefact from the richest man in England.
Tucci, who recently joined the cast of The Hunger Games, is best known for his Academy Award-nominated role in The Lovely Bones.
Leachman was well known in the '70s for portraying the character Phyllis Lindstrom on both The Mary Tyler Moore Show and spinoff series Phyllis.
Gambit began filming in London earlier this month, although CBS Films has yet to announce a release date.
