They join Gillian Anderson and Mark Williams in Matthew Butler's 18th century zombie comedy, Screen reports.
An extension of Butler's 2010 short E'gad Zombies, which also starred McKellen, the script has been co-written and produced by Butler with Tori Hart.
Margaret Matheson will executive produce. A budget of $3.3 million (£2 million) is being sought for the film, with plans to shoot in Scotland before the end of the year.
Last year, Dench was voted the greatest stage actor of all time, while McKellen was placed fourth in the same poll.
