The American football comedy - formerly known as Turkey Bowl - was first described as a project to reunite The Other Guys co-stars Wahlberg and Will Ferrell.
Speaking to Entertainment Weekly, McKay said: "Owen Burke, who's a producer at Gary Sanchez, and I were talking about how crazily funny Wahlberg and Baldwin were in The Departed - how, even though it's not a comedy, we could just watch them all day long.
"Burke dropped this idea, and I was like, 'Oh my God, that's a movie!' Then Will heard it and he wanted in."
Mississippi Three centres on a traditional Thanksgiving football game that pits rival families from across the street with each other.
Baldwin and Wahlberg will play father and son in the comedy, which McKay describes as an "ensemble" piece that will feature cameos from comedians in the style of Anchorman.
