Vaughn returned to the franchise to helm latest entry X-Men: First Class, after dropping out of X-Men: The Last Stand back in 2006.
The Kick-Ass director has since confirmed that he is definitely interested in working on a potential sequel should the new film prove a success.
"If the film's a hit I definitely want to work with Fox again," Vaughn told Superhero Hype. "There's a lot of fun things that can be done with this world.
"I'd be very interested in doing a sequel. The hard thing is creating a new franchise, so if it works it would be fun."
Vaughn even revealed that he had already planned out several ideas for a sequel to X-Men: First Class should Fox decide to move ahead with it.
"My ideas are much bigger for the next one," he added. "I'm surprised everyone is calling this a really big, epic move, and I'm thinking, 'Wait until you view the next one if you think this is epic, the next one you won't believe what happens'."
Rose Byrne, who portrays Moira McTaggert, recently claimed that X-Men: First Class was envisioned as the start of a whole new trilogy.
X-Men: First Class will launch in UK cinemas on June 1, and is released in the US two days later.
