The Superman Returns star will play a part-Native American coaching a lower-league lacrosse team, Variety reports.
His character will take up the call of duty from his tribe leader father by trying to steer the team to victory against a local prep school.
Routh told the site: "While it has all the ingredients of a classic underdog sports movie, it actually appealed to me on a deeper level.
"I think the father-son and brother-sister dynamics of the story are compelling, as is [the] more spiritual Native American aspect."
The actor was last seen in Dylan Dog: Dead of the Night, where he played a paranormal investigator.
He admitted last year before losing the role to Henry Cavill that he was keen to play Superman again.
