Mackie was previously linked to the project, and Variety reports that The Hurt Locker star has now officially signed up for the film.
The film is based on Paul Lieberman's seven-part Los Angeles Times series Tales From the Gangster Squad, about the LAPD's struggle with corruption during the forties.
Gangster Squad will also star Sean Penn, Ryan Gosling and No Country for Old Men lead Josh Brolin.
Mackie is also believed to have joined the cast of Marc Forster's World War Z in an unspecified role opposite Brad Pitt and Mireille Enos.
The actor will next be seen in the sci-fi sports drama Real Steel with Hugh Jackman.
