Van Ronk was a musician and a figurehead in the 1960s Greenwich Village music scene, who inspired several bigger names such as Joni Mitchell and Bob Dylan early in their careers.
The Coens' project would only be "loosely based" on the folk legend's life, which was related in the posthumous memoir The Mayor of McDougal Street, according to a source for the Los Angeles Times.
The two previously revealed that their follow-up to True Grit will feature mostly music "performed live, on a single instrument".
The film's naturalistic style is also reportedly likely to resemble Noah Baumbach's film Margot at the Wedding.
The directing duo recently worked on the screenplay for Michael Hoffman's remake of the 1966 caper Gambit, which stars Cameron Diaz, Colin Firth and Stanley Tucci among others.
