The Temple Grandin star will play the abolitionist William Seward, who served as a member of the president's war cabinet, Deadline reports.
Lincoln is based on the non-fiction account Team of Rivals by Doris Kearns, which centres on inter-cabinet rivalries ahead of the Civil War.
The actor will be joining confirmed cast members Daniel Day-Lewis and Sally Field, who will play the president and his wife Mary Todd Lincoln respectively.
Tommy Lee Jones was reportedly being considered for the role of anti-slavery campaigner Thaddeus Stevens, while James Spader and Joseph Gordon Levitt were also rumoured to be in negotiations.
Spielberg has two directorial efforts planned for release before Lincoln. His next two pictures - War Horse and The Adventures of Tintin: Secret of the Unicorn - are both scheduled to hit cinemas in December 2011.
