The actress, who will be seen in the next Twilight film as a Covenant vampire, is to play Benicio Del Toro's wife in the adaptation of Don Winslow's novel.
Her character will be "a tough and bitter woman whose soccer mum lifestyle hides a gang-life past", according to The Hollywood Reporter.
The film centres on a pair of drug dealers living in Laguna Beach, who are forced to work for a cartel after their shared girlfriend is kidnapped by the leader.
Maestro will join confirmed cast members Emile Hirsch, Blake Lively, Aaron Johnson, Uma Thurman and John Travolta.
The 33-year old Spanish actress made her Hollywood debut in Frida, the biopic of legendary Mexican artist Frida Kahlo.
