The 4 songstress announced her casting as aspiring singer Vicki Lester while performing on today's edition of Good Morning America, as part of the ABC show's summer concert series.
"Clint Eastwood? Can you believe that?" she gushed. "I can't believe it still. When I met with him I was just in awe. I'm so, so honoured that he believes in me. And I can't wait to do this film.
"I represent this generation's talent and for me to be the chosen person, as an African American woman, is incredible."
Beyoncé's selection for A Star Is Born had been rumoured since January, while Leonardo DiCaprio was recently touted to play the film's male lead.
A Star Is Born, which tells the story of an ageing movie star who helps a young actress find fame, was first released in 1937 and has been remade twice in 1954 and 1976.
Beyoncé's character has previously been portrayed by Janet Gaynor, Judy Garland and Barbra Streisand.
A Star Is Born is due to begin filming this autumn with a release date set for 2012.
