The Sherlock Holmes helmer is to replace Francis Lawrence in the director's chair, who originally took the reins from Ritchie in 2009, Twitch Film reports.
Sgt. Rock was created by Robert Kanigher and Joe Kubert, and published by DC Comics in 1959. The comic sees the character lead a fictitious World War II US Army infantry called Easy Company.
Ritchie is said to be overseeing the development of a new screenplay for the project, taking the adaptation away from Lawrence's proposed futuristic setting and back its World War II roots.
Joel Silver, who spent two decades trying to get the project greenlit, will produce alongside Akiva Goldsman. It was previously reported that Arnold Schwarzenegger was once linked with the title role.
Guy Ritchie's return to Sgt Rock is yet to be officially announced.
