The actor will be playing one half of a team of young policemen who are forced to join up with a female Delta Force operative in a mission to capture a mercenary.
Producer Joel Silver is behind the project, which has been described as an attempt at a Lethal Weapon-style franchise, Vulture reports.
Silver has previously produced the Mel Gibson and Danny Glover films, as well as other hits 48 Hours, The Matrix and Die Hard.
Running Scared director Wayne Kramer was originally attached to direct in 2009.
Timberlake will next be seen in comedy Friends with Benefits and Now with House star Olivia Wilde.
The star was recently given a personal invitation to the Marine Corps Ball by a female marine after he persuaded Friends with Benefits co-star Mila Kunis to visit the ball.
He told the actress that it was her patriotic duty to accept her invitation from the Marine Corps.
