According to The Hollywood Reporter, the two have been tapped to play a married couple in the project, formerly titled This is Forty, which serves as a spinoff from Apatow's 2007 comedy Knocked Up.
The project sees Paul Rudd and Leslie Mann reprise their roles as troubled couple Pete and Debbie opposite Bridesmaids actress Melissa McCarthy.
Mumolo co-wrote Bridesmaids with the film's lead actress Kristen Wiig and also had a small role in the film as a nervous flight passenger.
As well as working on Saturday Night Live's TV Funhouse shorts, Smigel is also well-known for both puppeteering and voicing Triumph, the Insult Comic Dog.
The currently-untitled Knocked Up spinoff is expected to reach cinemas in June 2012.
