The film follows the adventures of a non-Jewish plumber who pretends to be a doctor in order to woo a Jewish woman who will sort out his life for him.
Hewitt will play the woman he falls for at a Jewish singles night at a local temple, according to Variety.
The film has been described as a cross between My Big Fat Greek Wedding and Wedding Crashers.
Crossing Jordan star Ivan Sergei will play the male lead in the adaptation, written and directed by playwright Bryan Fogel.
His last big-screen appearance was in the 2006 romantic comedy The Break Up, with Jennifer Aniston.
Jewtopia became the longest-running original play in the history of Los Angeles theatre in 2003.
Hewitt was last seen in The Client List and The Lost Valentine.
She admitted that her mother wants her to appear in more wholesome, family-friendly films after she played a prostitute in the 2010 thriller.
In June, Hewitt was reportedly dropped from talks to play a recurring role in Law & Order: Special Victims Unit.
