The Black Swan actress, who welcomed her first child with fiancé Benjamin Millepied in June, was in talks for the lead part in the film Adaline.
However, a rep for Portman told Entertainment Weekly that she has turned down the offer.
Adaline centres on a woman who becomes ageless following an accident in the early 1900s. Years later, she meets a man for whom it may be worth forsaking her immortality.
Angela Lansbury has also been approached to star in the film.
Earlier this month, Portman insisted that she won't let the thought of failure stop her from doing something, saying that she knows "failing isn't that bad".
In May, the Oscar winner suggested that she may quit acting in favour of starting a family.
Portman previously confessed that she nearly suffered a "total breakdown" after completing production on Black Swan.
