He will be joining White Collar star Matthew Bomer, Matthew McConaughey, Alex Pettyfer and Channing Tatum, according to The Hollywood Reporter.
The film centres on Tatum's real-life experience as a teenage stripper with the G.I.Joe actor in the role of mentor to his young self, played by Pettyfer.
Iglesias, 35, has previously made guest appearances on Family Guy and chat shows such as The Late Late Show and Jimmy Kimmel Live.
He has also starred in independent feature films The Surfer King and El Matador.
Iglesias will play a DJ who works at the strip club run by Matthew McConaughey's former stripper character.
Cuban soap opera actor William Levy decided to turn down the project and was replaced by Iglesias.
Other casting additions could include True Blood star Joe Manganiello, who is reportedly in final talks for a role in the film.
Tatum recently claimed that he was "unashamed" of his past as an exotic dancer.
